Penelitian ini bertujuan untuk mengetahui keragaman genetik Andaliman (Zanthoxylum Acanthopodium D.C) di Sumatera Utara melalui analisis pola pita RAPD berdasarkan primer OPC 07, OPD 03, OPD 20, OPM 20, OPN 09. Penelitian ini telah dilakukan di Laboratorium Terpadu Fakultas Kedokteran USU pada April s/d September 2015 dengan menggunakan stok DNA dari 30 aksesi andaliman yang berasal dari tiga kabupaten di Sumatera Utara yaitu Kabupaten Dairi, Kabupaten Tanah Karo dan Kabupaten Simalungun. Total pola pita yang dihasilkan dari primer OPC07, OPD03, OPD20, OPM20, OPN09 adalah 20 pita yang 78.34% adalah pita polimorfik dan 21.66% pita monomorfik. Ukuran pita yang dihasilkan berkisar 418 bp – 2500 bp. Koefisien keragaman genetic dan dendogram filogenetik diperoleh menggunakan software Darwin 6.01 yang hanya dapat memproses 15 aksesi yang diuji karena sebagian aksesi tidak memenuhi persentase yang distandarkan. Analisis tersebut menunjukkan bahwa 15 aksesi andaliman tersebut terbagi dalam tiga kelompok. Pengelompokan aksesi andaliman tidak berdasarkan daerah, fisiologis tanaman dan ketinggian tempatnya. Penelitian ini menunjukkan bahwa 30 aksesi andaliman dari tiga kabupaten di Sumatera Utara memiliki keragaman genetik yang tinggi.
The aimed of the research was to determine the genetic diversity Sumatera’s andaliman based on OPD 03, OPD 20, OPC 07, OPM 20, OPN 09 primers. Five primers showed 20 band which is 78.34% polymorphic and 21.66% monomorphic. The size of DNA’s bands were varied from 418bp – 2500bp. The research was carried out in the Laboratorium Terpadu Fakultas Kedokteran University of Sumatera Utara began from April to September 2015 which used DNA stock from thirty accession of Andaliman from three regions in North Sumatera : Dairi, Karo and Simalungun. DARwin 6.01 software was used to calculate Factorial principal coordinates analysis and phylogenetic that only can process fifteen accessions because some of accessions have not software’s standart. The analysis showed that 15 accessions clustered into three groups. The clustering were not based on region, plant physiology and altitude. The research showed that the thirty accession of andaliman from three regions in North Sumatera has a high genetic diversity.
