Analysis of PCR Amplification of Date Palm (Pheonix dactylifera L.) Genotypes Based on RAPD Primers

Date palm cultivation can be done using seeds, offshoot and through tissue culture (in vitro). Propagation through tissue culture also has disadvantages such as genetic changes known as somaclonal variation. The use of molecular markers can help early detection of genetic variation. One type of molecular marker commonly used for genetic variation is RAPD (Random Polymorphic Amplified DNA) markers. The aim of the research was to analyze the polymorphism of Date Palm genotypes based on three RAPD primers. This research is conducted by taking 8 samples from tissue culture propagation and 2 samples from seed propagation. A total of 15 bands were scored, with the range of band size was 214bp - 1918bp measured using UVITEC Cambridge FineReader software and the average of polymorphism was 74.40%. The absorbance value of DNA stock in Å260 / 280 ranged from 1.96 - 2.46
